Well, yes and no.
You can play with other people over the internet using WiiConnect24, but as austryhard said, no games have been released yet that support that feature.
And even then, it's not going to be like XBox Live!. As far as we've heard there is no matchmaking or anonymous play — you can only play games with Wii Friends you have added to your address book. This is because Nintendo, for some crazy reason, has decided to stick with the DS's system of Friend Codes instead of adopting an open server solution like other developers have been doing *since the first MUD in 1979*.
